Building a vessel wall with notch signaling.

چکیده
Blood vessel walls are built and maintained by 2 interdependent cell types, endothelial cells and mural cells. Signals exchanged between these 2 cell types control the formation, maturation, remodeling, and function of microvascular networks. Mural cells are either pericytes in the microvasculature or smooth muscle cells (SMCs) in larger vessels. Interactions between endothelial cells and mural cells serve to stabilize nascent capillary vessels, provide endothelial cell survival factors, inhibit endothelial cell proliferation, promote mural cell differentiation, and guide vascular network remodeling.1,2 Yet precisely how endothelial cells recruit mural cells to form a functional vessel wall is still incompletely understood.

Differential Notch signaling in the epicardium is required for cardiac inflow development and coronary vessel morphogenesis.
RATIONALE The proepicardium is a transient structure comprising epicardial progenitor cells located at the posterior limit of the embryonic cardiac inflow. A network of signals regulates proepicardial cell fate and defines myocardial and nonmyocardial domains at the venous pole of the heart. During cardiac development, epicardial-derived cells also contribute to coronary vessel morphogenesis. ...
متن کاملBlocking the NOTCH pathway inhibits vascular inflammation in large-vessel vasculitis.
BACKGROUND Giant cell arteritis is a granulomatous vasculitis of the aorta and its branches that causes blindness, stroke, and aortic aneurysm. CD4 T cells are key pathogenic regulators, instructed by vessel wall dendritic cells to differentiate into vasculitic T cells. The unique pathways driving this dendritic cell-T-cell interaction are incompletely understood, but may provide novel therapeu...
متن کاملVon Willebrand Factor Inhibits Mature Smooth Muscle Gene Expression through Impairment of Notch Signaling
Von Willebrand factor (vWF), a hemostatic protein normally synthesized and stored by endothelial cells and platelets, has been localized beyond the endothelium in vascular disease states. Previous studies have implicated potential non-hemostatic functions of vWF, but signaling mechanisms underlying its effects are currently undefined. We present evidence that vWF breaches the endothelium and is...

متن کاملNotch controls endothelial cells
Notch signaling is an evolutionarily conserved pathway that controls numerous cell differentiation steps during development. Also in the adult, Notch signaling is essential to control stem cell differentiation in the bone marrow or the gut mucosa. Here, we briefly emphasize the roles of Notch during development of blood vessels (angiogenesis). Further, we highlight novel findings that indicate ...
